How can I have "letter/block in own column" selected by default? There is an Input for the TPO Profile named Default Column Display Style (for All Profiles). You will want to set this to Letters/Blocks in Own Column. Then you need to save the Inputs as they are at that moment as your Default Settings for that study by selecting the button at the bottom for Set Defaults.
