You do not need to use the Define Custom Colors option. Once the color selection window is open, select one of the 16 Custom Colors boxes and then adjust the color on the right to be what you want. Then select the Add to Custom Colors button and it will update that specific Custom Color box. This is all described in the documentation referenced above.
