I have set the Trade Position Limit setting for a symbol to a non-zero value but it seems to have no effect in either simulation or live trading, as I am able to place a new order of larger quantity than the limit without rejection. This was for symbol MNQU23_FUT_CME with Teton CME Order Routing. Is there some other setting I need to change as well to activate the Trade Position Limit?

We just tried this to ensure that it is working properly, and it is working as expected.

The most likely item is that you do not have the option for "Use Custom Symbol Settings Values" in the top of the Symbol Settings window, enabled. Make sure this has a checkmark next to it. If not, then any custom settings that you make for the symbols will not be used. Refer to the following:
Global Symbol Settings: Use Custom Symbol Settings Values (Global Settings >> Symbol Settings)

I see in the docs that if you set Trade Position Limit to 0 in Symbol Settings - that means there is no limit. Is there a way to actually limit the number of contracts to 0?

There is not a way to limit the number of contracts to 0.

If you do not want to trade, then enable the option for "Trade >> Trading Locked". Refer to the following:
Trade Menu: Trading Locked (Trade menu)
For the most reliable, advanced, and zero cost futures order routing, use the Teton service:
