This question is not clear enough. Where is it in Sierra Chart that you are looking at the profit/loss of a Trade?

You first need to specify the commission amount properly in the Global Symbol Settings for the symbol pattern.

And then you can enable the use of the Commission setting for the Trades list. This is all documented here:
Chart Settings: Use Symbol Commission Setting in Trade List and Statistics Calculations (Chart >> Chart Settings >> Trading menu)

Is it possible to specify the fees in more detail?
For example for a trade based on the Maker / Taker model, the fee for Limit orders is different from Market orders. In the Global Symbol settings it is not possible to break fees down in such detail. A round turn could be any combination of Maker (limit, stop limit) or Taker (market, stop market) orders, the stats on this being available, it should be possible to calculate the PnL more accurately. Percentage terms entry is required.
