Sierra Chart will run, but we do not recommend doing what you are suggesting. In fact, due to the amount of data that we read from disk, we recommend using Solid State Disks internal to the machine. When you connect an external drive, you are then going over the USB interface, which is much slower than the SATA interface that is internal to your system. The best thing would be to upgrade the drive in your computer to an NVMe SSD drive and then reinstall Sierra Chart there.
